Assessment of 5-Azacitidine Efficacy in Cisplatin-Resistant Germ Cell Tumor Models
Introdução
Germ cell tumors (GCTs) are neoplasms from primordial germ cells, which can be benign or malignant. The treatment typically involves surgery and cisplatin-based chemotherapy, which binds to DNA, leading to cell death. However, resistance to cisplatin is a significant challenge in some patients. 5-azacitidine was approved by the FDA for treating pediatric and young adult leukemia. It is hypothesized that 5-azacitidine could also be effective against GCTs by inducing genomic hypomethylation and loss of epigenetic memory.
Objetivo
To evaluate the effect of 5-azacitidine treatment in an animal model of cisplatin-resistant GCTs.
Métodos
Athymic nude mice (NU/J strain) aged eight weeks from the CPOM-HCB animal facility will be used. GCT cell lines, both NTERA-2 parental and cisplatin-resistant, will be implanted subcutaneously. After tumor formation, treatments with 5-azacitidine and cisplatin will commence. Tumor progression will be monitored, and histological and DNA methylation analyses will be performed. This project has been approved by the CEUA under protocol number 031/2023.
Resultados
It is expected that 5-azacitidine will induce DNA hypomethylation in tumor cells, restoring their sensitivity to cisplatin and resulting in tumor volume reduction.
Conclusões
The completion of this work is essential, as the expected results could pave the way for new therapeutic strategies.
