Obatoclax Mesylate effects on cell morphology and death in Triple Negative Breast cancer cells (TNBC)
Introdução
Triple negative breast cancer (TNBC) is highly aggressive and is characterized by poor prognosis. Apoptosis and autophagy are mechanisms responsible for maintaining cellular homeostasis. When deregulated, they can promote cancer progression. The synthetic compound Obatoclax Mesylate (OBX) acts by binding to anti-apoptotic proteins, therefore allowing the pro-apoptotic proteins to initiate the apoptosis pathway. Recent studies demonstrated apoptotsis and autophagy induction in several types of cancer treated with this compound.
Objetivo
Considering the lack of effective therapies for TNBC, it becomes relevant to extend the studies of OBX in this type of cancer.
Métodos
The triple negative metastatic human Hs578T breast cancer cell line was used in the present study. Initially, the IC50 concentration of OBX and growth curve were determinated by using Trypan Blue exclusion test. Then, cells were treated with OBX and the morphology was analyzed by fluorescent staining of cells with phalloidin-Alexa555, and immunofluorescent reaction with anti-Tubulin antibody. Additionally, a time-lapse assay was done, to analyze the effects of OBX in cell morphology along time, for 20 hours.
Resultados
OBX decreased Hs578T cell viability, modified cell morphology and the microfilament organization to a more compacted one, which may reduce migration capacity. Furthermore, with the time-lapse, it was possible to observe the time point in which cells start to change morphologically, along with vacuoles formation, cytokinetic and adhesion modification and the continuity of mitosis after treatment.
Conclusões
The results of the present research could indicate an impairment of cell adhesion and the metastatic ability by action of OBX on Hs578-T cells. Although the cell viability assay showed cell death, more tests need to be done to confirm this effect.
